Cryosectioning and Immuno-Electron Microscopy

Meeting
Date: 2024-03-11 to 2024-03-15

This 4 day workshop (March 11-15, 2024) is organized by our Correlative Light Microscopy Dutch Flagship Node in collaboration with Leica Microsystems and is meant for people that do not have experience with the Tokuyasu technique. Experience in resin sectioning is not necessary, but is definitely an advantage. The workshop is primarily practical in nature, although some topics will be dealt with in lectures. The entire procedure from (live) specimen till immuno-labeled ultrathin section will be covered from start to finish. Most of the time will be spent in the lab working with standard specimens like cultured cells or tissues. Besides managing the ultra cryo-microtome, the preparation of tools and other necessities will be covered.

We provide an excellent learning environment. When manual skills like ultrathin sectioning are being trained the participants will have their own ultra-cryo microtome (often equipped with a camera) to practice on. The number of participants is limited to 12 people. We will provide you with a practical manual covering the topics of the course.
A preliminary program and a more elaborate list of topics that are addressed in this course can be found here.

Available equipment: 2 x Reichert Ultracut E, 3x Leica Ultracut S, 2x Leica Ultracut T, 1x Leica UC6/FC6 ,1 x Leica UC6, 2x Leica UC7. Leica KMR3, Leica AC20, Leica EM AMW, Leica EM TRIM2, Leica EM ACE600, Edwards Auto 306 carbon evaporator, JEOL electron microscopes, 2 FEI electron microscopes.
