Mina Shiran Chaharsoughi, Ph.D, from Exciscope will present on "Non-destructive 3D histology with x-ray phase-contrast tomography" in our first Tech Exchange episode after the summer break.

Abstract:

Histological analysis by light microscopy has long been the gold standard for biological soft tissues and other low-density samples. While the resolution is high – down to 200 nm – it is a highly invasive procedure where the sample preparation involves fixation, dehydration, embedding, sectioning and staining. The process of slicing the sample effectively limits the imaging to two dimensions. With our instrument, the Exciscope Polaris, biological or other low-density samples can be scanned to render a 3D volume with 1 um resolution. In this presentation, we will introduce the x-ray phase-contrast technique we use, give an overview of the specialised hardware and software needed to achieve it, and last but not least, show what results you can expect from a range of different samples, for example soft-tissue tumours, plants, and ancient human remains.
